Skin health: how oral supplementation supports skin barrier function, dermal density, and signs of aging

Skin aging is driven by a combination of intrinsic and environmental factors that progressively affect both the epidermis and dermis. These changes can lead to impaired barrier function, reduced elasticity and firmness, loss of dermal density, and the appearance of wrinkles.

A recent open-label clinical study investigated the effects of oral supplementation with a dry aqueous extract of Brassica oleracea standardized in glucoraphanin on instrumental and visible markers of skin aging.

The 56-day study included 50 healthy women aged 35–55 years with mild-to-moderate signs of aging. Participants consumed one tablet daily and underwent objective skin assessments using validated non-invasive instrumental methods.

Key outcomes after 56 days

  • Improved skin barrier function
    • Transepidermal water loss (TEWL) decreased by 7.5%
    • Stratum corneum cohesivity improved, with 27.8% lower protein removal
  • Enhanced dermal structure
    • Dermis density increased by 12.8%
  • Visible anti-aging effects
    • Wrinkle depth decreased by 15.1%
    • Skin firmness improved by 17.4%
    • Skin elasticity increased by 9.7%

Participants also reported high levels of satisfaction regarding overall skin appearance and perceived skin quality.

The proposed biological mechanism is linked to glucoraphanin, a naturally occurring glucosinolate in Brassica oleracea, which can be converted by the gut microbiota into sulforaphane. Sulforaphane has been associated with activation of the NRF2 pathway, a key regulator involved in antioxidant defense and maintenance of skin homeostasis.

Overall, the study suggests that oral supplementation with Brassica oleracea extract may represent a promising inside-out strategy to support healthy skin aging, helping preserve both barrier integrity and dermal structure.
